A dynamic and mechanistic model of PCB bioaccumulation in the European hake (Merluccius merluccius) ArchiMer
Bodiguel, Xavier; Maury, Olivier; Mellon-duval, Capucine; Roupsard, Francois; Le Guellec, Anne-marie; Loizeau, Veronique.
Bioaccumulation is difficult to document because responses differ among chemical compounds, with environmental conditions, and physiological processes characteristic of each species. We use a mechanistic model, based on the Dynamic Energy Budget (DEB) theory, to take into account this complexity and study factors impacting accumulation of organic pollutants in fish through ontogeny. The bioaccumulation model proposed is a comprehensive approach that relates evolution of hake PCB contamination to physiological information about the fish, such as diet, metabolism, reserve and reproduction status. The species studied is the European hake (Merluccius merluccius, L. 1758). The model is applied to study the total concentration and the lipid normalised...

A multitracer approach to assess the spatial contamination pattern of hake (Merluccius merluccius) in the French Mediterranean ArchiMer
Cresson, Pierre; Bouchoucha, Marc; Morat, Fabien; Miralles, Francoise; Chavanon, Fabienne; Loizeau, Veronique; Cossa, Daniel.
Chemical contamination levels and stable isotope ratios provide integrated information about contaminant exposure, trophic position and also biological and environmental influences on marine organisms. By combining these approaches with otolith shape analyses, the aim of the present study was to document the spatial variability of Hg and PCB contamination of the European hake (Merluccius merluccius) in the French Mediterranean, hypothesizing that local contaminant sources, environmental conditions and biological specificities lead to site-specific contamination patterns. High Hg concentrations discriminated Corsica (average: 1.36 ± 0.80 μg g− 1 dm) from the Gulf of Lions (average values < 0.5 μg g− 1 dm), where Rhône River input caused high PCB burdens....

A steady-state model of PCB accumulation in dab food web ArchiMer
Loizeau, Veronique; Menesguen, Alain.
A five-compartment steady-state food web model is proposed for the benthic food web leading to the dab. Three exposure pathways are considered in the description of accumulation by benthic animals: ingestion of particulate contaminants associated with either sediment or phytoplankton, and respiratory uptake of free dissolved contaminant in overlaying water. Application of the model to a simple food web in the Bay of Seine (Eastern Channel) indicates that : a) feeding is the principal route of contamination, especially for PCB which have more than four chlorine atoms in the molecule ; b) excretion and growth rates, phytoplankton lipid fraction and organic carbon content of sediment are the parameters which mostly determine the chemical bioaccumulation in...

Comparison of C and N stable isotope ratios between surface particulate organic matter and microphytoplankton in the Gulf of Lions (NW Mediterranean) ArchiMer
Harmelin Vivien, M; Loizeau, Veronique; Mellon-duval, Capucine; Beker, B; Arlhac, D; Bodiguel, Xavier; Ferraton, Franck; Hermand, R; Philippon, Xavier; Salen Picard, C.
Carbon and nitrogen stable isotope ratios of particulate organic matter (POM) in surface water and 63-200 mu m-sized microphytoplankton collected at the fluorescence maximum were studied in four sites in the Gulf of Lions (NW Mediterranean), a marine area influenced by the Rhone River inputs, in May and November 2004. Some environmental (temperature, salinity) and biological (POM, Chlorophyll a and phaeopigments contents, phytoplankton biomass and composition) parameters were also analysed. Significantly different C and N isotopic signatures between surface water POM and microphytoplankton were recorded in all sites and seasons. Surface water POM presented systematically lower delta C-13 (similar to 4.2 parts per thousand) and higher delta N-15 (similar to...

Differential biomagnification of PCB, PBDE, Hg and Radiocesium in the food web of the European hake from the NW Mediterranean ArchiMer
Harmelin-vivien, Mireille; Bodiguel, Xavier; Charmasson, Sabine; Loizeau, Veronique; Mellon, Capucine; Tronczynski, Jacek; Cossa, Daniel.
Consumption of marine organisms represents one of the main exposure sources of contaminants for human populations. To obtain a global view of the contamination in commercial fish in the NW Mediterranean Sea, we analysed four types of priority contaminants (PCBs, PBDEs, Hg and Cs-137) in the European hake, Merluccius merluccius, from the Gulf of Lions in relation with organism's trophic level (delta N-15). All contaminants presented a significant increase in concentration in hake muscle with trophic level. However, obvious differences between contaminants were evidenced. Biomagnification factors (BMF and FWMF) along the hake food web were higher for Hg and CB-153 than for BDE-47 and Cs-137, and increase in contaminant concentration with trophic level...

Fish life-history traits are affected after chronic dietary exposure to an environmentally realistic marine mixture of PCBs and PBDEs ArchiMer
Horri, Khaled; Alfonso, Sebastien; Cousin, Xavier; Munschy, Catherine; Loizeau, Veronique; Aroua, Salima; Begout, Marie-laure; Ernande, Bruno.
Polychlorinated biphenyls (PCBs) and polybrominated diphenyl ethers (PBDEs) are persistent organic pollutants that have been shown to affect fish life-history traits such as reproductive success, growth and survival. At the individual level, their toxicity and underlying mechanisms of action have been studied through experimental exposure. However, the number of experimental studies approaching marine environmental situations is scarce, i.e., in most cases, individuals are exposed to either single congeners, or single types of molecules, or high concentrations, so that results can hardly be transposed to natural populations. In the present study, we evaluated the effect of chronic dietary exposure to an environmentally realistic marine mixture of PCB and...

Influence of sex, maturity and reproduction on PCB and p,p ' DDE concentrations and repartitions in the European hake (Merluccius merluccius, L.) from the Gulf of Lions (NW Mediterranean) ArchiMer
Bodiguel, Xavier; Loizeau, Veronique; Le Guellec, Anne-marie; Roupsard, Francois; Philippon, Xavier; Mellon-duval, Capucine.
The main objective of this work was to establish the influence of sex, maturity and reproduction on the contamination of the demersal fish Merluccius merluccius by organochlorine compounds. Polychlorinated biphenyls (PCBs) and p,p'DDE were quantified in muscle, liver and gonads of female and male hakes collected in the Gulf of Lions in 2004 and 2005. Observed levels appeared higher than the population of the Bay of Biscay and lower than the population of the Thyrrenian Sea. Contaminant fingerprints were roughly constant whatever the studied organ and the hake biological condition. Concentrations varied significantly according to the sex and maturity of hakes. Mature specimens were more contaminated than immature, and males presented higher levels than...

Long-term dietary-exposure to non-coplanar PCBs induces behavioral disruptions in adult zebrafish and their offspring ArchiMer
Pean, Samuel; Daouk, Tarek; Vignet, Caroline; Lyphout, Laura; Leguay, Didier; Loizeau, Veronique; Begout, Marie-laure; Cousin, Xavier.
The use of polychlorinated biphenyls (PCBs) has been banned for several decades. PCBs have a long biological half-life and high liposolubility which leads to their bioaccumulation and biomagnification through food chains over a wide range of trophic levels. Exposure can lead to changes in animal physiology and behavior and has been demonstrated in both experimental and field analyses. There are also potential risks to high trophic level predators, including humans. A maternal transfer has been demonstrated in fish as PCBs bind to lipids in eggs. In this study, behavioral traits (exploration and free swimming, with or without challenges) of contaminated zebrafish (Danio rerio) adults and their offspring (both as five-day-old larvae and as two-month-old fish...

Long-term exposure to polybrominated diphenyl ethers (PBDEs) or polychlorinated biphenyls (PCBs) through diet affects embryonic development of common sole (Solea solea) ArchiMer
Omnes, Marie-helene; Lemerre, Charlotte; Le Bayon, Nicolas; Le Delliou, Herve; Zambonino, Jose-luis; Munschy, Catherine; Loizeau, Veronique.
Persistent organic pollutants (POPs), such as PCBs (polychlorinated biphenyls) and PBDEs (polybrominated diphenyl ethers), are lipophilic and tend to bioaccumulate in cellular membranes and fat. The accumulation of lipids in oocytes during vitellogenesis, and subsequently in the eggs, favors the accumulation of POPs, and may affect the development of embryos. The effects of dietary exposure to POPs in Common sole Solea solea were evaluated on early life stages (ELS): the survival of sole embryos and features of embryonic development were assessed until hatching endpoint and compared to that of the control. No embryonic development was observed in PBDE eggs while hatching occurred in both control and PCB groups. In addition, several developmental...

Long-term food-exposure of zebrafish to PCB mixtures mimicking some environmental situations induces ovary pathology and impairs reproduction ability ArchiMer
Daouk, Tarek; Larcher, Thibaut; Roupsard, Francois; Lyphout, Laura; Rigaud, Cyril; Ledevin, Mireille; Loizeau, Veronique; Cousin, Xavier.
Although the use of polychlorinated biphenyls (PCBs) has been banned for several decades, they are still present in the environment and are occasionally mechanically released from sediment or transferred through the trophic chain. Field analyses have established correlations between exposure to PCBs and alterations in fish physiology including reproductive function. Experimental exposures have been mainly performed using dioxin-like PCBs or other congeners at very high concentrations. However, these studies are often difficult to relate to real-life conditions. In the present study, we performed a life-cycle exposure using zebrafish model and mixtures representative of some environmental situations in terms of doses, composition and containing mainly non...

Modelling growth and bioaccumulation of Polychlorinated biphenyls in common sole (Solea solea) ArchiMer
Eichinger, Marie; Loizeau, Veronique; Roupsard, Francois; Le Guellec, Anne-marie; Bacher, Cedric.
Experiments were performed on juvenile sole in controlled conditions in the aim of understanding how the biology of common sole may affect the accumulation and dilution of Polychlorinated biphenyls (PCBs). The fish were raised in optimal conditions and divided into two tanks: one control tank and one PCB tank. 4 PCB congeners were added to food for 3 months in the PCB tank; the soles were subsequently fed unspiked food for 3 months. Growth (length and weight) and PCB concentrations were monitored in both tanks and juvenile sole growth was not significantly affected by PCBs in our experimental conditions. We used the Dynamic Energy Budget (DEB) theory to model sole biology and paid special attention to model calibration through the wide use of data from the...

Organochlorinated contaminants in decapod crustaceans from the coasts of Brittany and Normandy (France) ArchiMer
Bodin, Nathalie; Abarnou, Alain; Le Guellec, Anne-marie; Loizeau, Veronique; Philippon, Xavier.
The contamination and distribution of organ ochlorinated compounds were considered in three crustacean species (edible crab, Cancer pagurus; spider crab, Maja brachydactyla; velvet swimming crab, Necora puber) from five sites along the coasts of Brittany and Normandy (Western and North-Western France). PCBs (16 single congeners), pp'-DDE and HCB were measured in hepatopancreas, gonads and muscle: in all, 175 samples were analysed. The spider crab was the only species found in the five sampling sites, thus enabling comparison between areas. Specimens from Antifer were much more contaminated (Sigma(16)PCBs in hepatopancreas = 2000-4000 ng g(-1) dry weight) than those from other sites (50-1000 ng g(-1) d.w.). Among all the three species, the spider crab...

PCB, PCDD/F and PBDE levels and profiles in crustaceans from the coastal waters of Brittany and Normandy (France) ArchiMer
Bodin, Nathalie; Abarnou, Alain; Fraisse, D; Defour, S; Loizeau, Veronique; Le Guellec, Anne-marie; Philippon, Xavier.
Polychlorinated biphenyls (PCBs), polybrominated diphenyl ethers (PBDEs) and polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs) were analysed in the muscle of various edible marine crustaceans (spider crab, edible crab, velvet swimming crab and Norway lobster) from the Brittany and Normandy coasts (France). The highest concentrations were measured in species collected from Antifer (Seine Bay). PCB and PBDE patterns in crustacean muscles were similar and independent of the geographical area with the predominance of the high chlorinated PCBs (CB 15 3, 138, 118 and 180), and of a few PBDE congeners (BDE47, BDE99, WE 100 and BDE28). Oppositely, dioxin contamination differed with site. The major component in crustaceans from the Seine Bay was...

Polycylic aromatic hydrocarbons (PAHs) are ubiquitous contaminants in the marine environment Their toxicity is mainly linked to the ability of marine species to biotransform them into reactive metabolites PAHs are thus often detected at trace levels in animal tissues For biomonitoring purposes this findings have two main consequences (i) the determination of the PAH tissue concentration is not suitable for the evaluation of individual exposure to PAHs (ii) It can explain sometimes the lack of correlations obtained with relevant markers of toxicity such as genotoxicity biomarkers The aim of the present study was to better investigate the link between PAH exposure and genotoxicity in marine flatfish During a laboratory experiment juvenile soles were exposed...

Responses of European anchovy vital rates and population growth to environmental fluctuations: An individual-based modeling approach ArchiMer
Pethybridge, Heidi; Roos, David; Loizeau, Veronique; Pecquerie, Laure; Bacher, Cedric.
A size-structured, bioenergetics model was implemented to examine the effects of short-term environmental changes on European anchovy, Engraulis encrasicolus, in the North-western Mediterranean Sea. The model approach was based on Dynamic Energy Budget (DEB) theory and details the acquisition and allocation of energy (J d(-1)) during an organisms' full life-cycle. Model calibration was achieved using biometric data collected from the Gulf of Lions between 2002 and 2011. Bioenergetics simulations successfully captured ontogenetic and seasonal growth patterns, including active growth in spring/summer, loss of mass in autumn/winter and the timing and amplitude of multi-batch spawning events. Scenario analysis determined that vital rates (growth and fecundity)...
